First published Friday, September 11, 2015 The University awarded a contract for 6550iFunnel Q-TOF LC/MS System to Agilent Technologies, Incorporated, Santa Clara, CA, for an estimated $445,895. Purchase from Agilent it's 6550iFunnel Q-TOF LC/MS System to be utilized for research. The principle investigator has previous experience with the Agilent instrumentation and software platform. Acquisition of this system will lead to reduced time spent on learning new software and additional training that would be necessary given other vendor platform acqusition. Recent improvements in acquistion and data analysis software provide a user-friendly interface to acquire data and process that data in a seemless manner. Furthermore, the software is "self-contained" such that additional purchases are not necessary for data processing. Mass Hunter is the software platform that will be used and it integrates into Spectrum Mill which will provide additional analysis resources. The MassHunter Software is required to operate both the chromatography system and the mass spectrometer. Without this software, the instrument can not be used. Spectrum Mill is a software for data interpretation. Therefore, it is also necessary as any data collected from the mass spectrometer will be imported for analysis. Without this piece of software, the data will be un-interpretable. It should be noted that a single data collection experiment will generate file sizes of approximately 1GB. Therefore, there is a lot of data incorporated in these files. One could not manually attempt to interpret. There are multiple factors that we have to consider for the instrument, some being more important than others and we understand there is always trade-offs: speed, resolution, mass accuracy, sensitivity as examples. For the research being conducted in the group, we need an instrument that has good resolution (termed high resolution) however, we do not need ultra-high resolution. Therefore, TOF mass analyzers are preferred over ICR or Orbitrap analyzers which are also considered high-resolution but capabilities are beyond the need. These analyzers are also slow, so the TOF analyzer is faster at data collection than the other two mentioned. The performance metrics of the Agilent 6550 in terms of resolution are >40,000 resolving power. At this level, we could easily differentiate common compounds that have similar masses, but differ at the hundredth's place, using 1000 as an example mass. Related, the scan speeds are 50 spectra/sec for data acquisition. For quantitative studies in the group, we need the instrument to perform in a linear fashion over several orders of magnitude to accommodate the variable concentrations of compounds in our samples. The Agilent 6550 provides linearity over 5 orders of magnitude. The only types of mass spectrometers which are better, are a different configuration which would not suit our needs and provide low resolution. Sensitivity and limits of detection metrics are important given that we are interested in compounds at low levels (ng/mL) and some of our samples are volume limited. For those that are limited, we will use the nanospray ion source and limits of detection are in the attomole range. Another feature is that the JetStream source provides a 5-10x increase in signal compared to standard ESI. For our samples which are better suited for standard ESI, this means that we will be able to use 5-10X less material than we would for standard ESI. This technology is only provided by Agilent. Furthermore, the sensitivity is improved via the dual iFunnel technology and hexabore capillary. These features are only available on the Agilent systems. Finally, with regard to mass accuracy, we are performing studies in which we need to confidently identify compounds in biological matrices related to human disease. Mass accuracy is crucial to this. The Agilent 6550 provides excellent, <2ppm mass error for a variety of experiments.
