The National Institute of Child Health and Human Development has a requirement to provide laboratory analysis of 6500 serum and 6500 urine specimens obtained from pregnant women during a previous Study. Analysis of serum specimens will be for soluble fms-like tyrosine kinase 1 (sFlt1 or sVEGFR1), free placental growth factor (PGF or PlGF), and free vascular endothelial growth factor A (VEGF), using enzyme-linked immunoassay (ELISA) kits obtained from R&D Systems Inc. (Minneapolis, MN in order to be consistent with other analysis undertaken by NICHD) as directed by NICHD. VEGF analyses must use an assay as sensitive as possible in order to be able to provide useful measurements of serum concentrations as low as 1 pg/ml or lower. Analysis of urine specimens will be for free PGF and for creatinine. Results will be entered on an Excel spreadsheet provided by NICHD, or as otherwise directed by NICHD. Requirements include significant experience performing ELISAs involving thousands of specimens; in depth knowledge of the particular ELISAs to be used; experience in conducting high sensitivity ELISAs for VEGF, capable of detecting 1 pg/ml or less; ability to confirm the validity of the urinary PGF ELISA; and the presence of experienced, skilled technical support to troubleshoot and monitor the performance of the ELISAs. All ELISAs to be performed by no more than two different laboratory technicians in order to maintain consistency of the laboratory results.
