There is only one drug that has been approved by the FDA for treatment of sickle cell disease. The drug is only partially successful, so there is an urgent need for addtional drugs. The Eaton group of 8 PhD scientists and one post-bac are working full time to discover an additional drug using a high throughput drug screen on intact sickle cells. The screen measures sickling times and judges the therapeutic potential of a candidate drug by its increase in sickling times, which is widely distributed because of the wide distribution of intracellular hemoglobin concentrations. The sickling time is enormously sensitive to the intracellular hemoglobin concentration, so knowing the distribution of intracellular concentrations, which is different for every patient, is critical for understanding the results of the screen. All hematology systems only measure the mean intracellular hemoglobin concentration, which is insufficient because of the wide and often bimodal distribution for sickle cells. Thus we require the entire distribution, which requires that the instrument measures and reports the intracellular hemoglobin concentration for each of the thousands of cells that are analyzed.
We urgently need an instrument that can measure the distribution of intracellular
hemoglobin concentrations. The Advia2120 Hematology System of Siemens
Healthineers is the only instrument capable of making these measurements. The Advia 2120 can do this because, unlike every other hematology instrument, it utilizes a dualangle light scattering method which allows determination of both volume and hemoglobin concentration of every cell. There is no competing instrument that has this capability.
