Background. The NCI Office of Cancer Genomics (OCG) sponsors two major initiatives that require cDNA sequencing support. They are the Cancer Genome Anatomy Project (CGAP) and the Mammalian Gene Collection (MGC). Sequencing support requirements include 5â and 3â sequencing of cDNA clones, and sequencing of SAGE library clones. Both sequencing requirements support downstream bioinformatics analyses to characterize gene expression and to select full ORF cDNA clones.
The NCI Cancer Genome Anatomy Project (CGAP) provides data that includes gene expression profiles of normal, precancer, and cancer cells based on EST and SAGE tags, SNP analysis of cancer-related genes, and chromosomal aberrations. Biological resources and the sequences generated by the project are available to the scientific community and the CGAP web site (cgap.nci.nih.gov) provides access to all CGAP data and analysis tools.
The trans-NIH Mammalian Gene Collection (MGC, mgc.nci.nih.gov) provides a comprehensive set of full-length (open reading frame, ORF) sequences and cDNA clones for expressed genes from human and mouse, and a partial set for rat. In addition, the OCG/NCI and the NHGRI are involved in the management of two other trans-NIH full-length ORF projects: Xenopus Gene Collection (XGC, xgc.nci.nih.gov) and Zebrafish Gene Collection (ZGC, zgc.nci.nih.gov).
