USDA, Agricultural Research Service, Pullman, WA, requires bi-directional sequencing of 7,680 clones, using the following methods: colony picking of inoculation of clones from 96 or 384 glycerol plates, cell culturing and DNA purification using SPRI, dye-terminator fluorescent sequencing reaction set-up, sequencing product purification using CleanSEQ, detection of sequencing fragments via capillary electrophoresis, creation of chromotograms, PHRED quality scores, and FASTA sequence files, and data transfer via secured FTP site.
