The specialized reagents are required to allow our staff at the NICHD to isolate RNA from single cells and to generate cDNA libraries from this RNA. Most importantly, these reagents will allow our staff to tag each RNA so that it is only counted once in our downstream analyses. These processes required highly specialized reagents and highly specialized and expensive equipment. Specifically, our staff need to use the 10xGenomics Microfluids Platform already available on campus. This machine requires 10xGenomics reagents enough for 20 reactions. This is because our staff need to generate enough statistical power to answer the question of whether RNAs from genes expressed at low levels are expressed from one parental allele or from both in each cell.
In summary, our staff must use single cell RNA sequencing with uniquely tagged RNAs for our study. Based on our consultations with experts, we agree that the 10xGenomics Microfluids Platform is the best system for our study and moreover it is the only system available to us. Only 10xGenomics reagents work on this Platform.
The 10xGenomics Chromium system will enable us to discover otherwise inaccessible genomic information and specifically will allow us to determine the parental origin of each RNA species from a single cell. Proprietary microfluid circuits rapidly partition single cell input samples, which are then barcoded using GemCode proprietary technology. The system has a library of up to 4,000,000 available barcodes which is necessary for us to analyze 1000's of RNAs from 1000's of cells. As described above, we will use a Platform already available on campus. We need to purchase the reagents to generate the single cell cDNA libraries that will be compatible with this platform.
