REQUEST FOR INFORMATION

 

1.0 RFI TITLE: IN VITRO Absorption, distribution, metabolism, and excretion (ADME) CHEMICAL SCREENING support services

2.0 AGENCY: U.S. Army Medical Reserarch Acquisition Activity (USAMRAA)

3.0 OFFICE: U.S. Army Medical Research Institute of Chemical Defense (USAMRICD).

4.0 LOCATION: Ft. Detrick, MD

5.0 SUBJECT: USAMRICD is conducting market research to determine whether a commercial solution can be provided to meet requirements for the Chemical Screening Support Services.  This market research may also assist with the further development and/or refinement of the requirements.

This RFI is issued solely for information and planning purposes and does not constitute a solicitation. Neither unsolicited proposals nor any other kind of offers will be considered in response to this RFI. Responses to this notice are not offers and will not be accepted by the Government to form a binding contract. In keeping with the standards of FAR Part 10, the Government may request additional information from responders. Responders are solely responsible for all expenses associated with responding to this RFI or any follow-up information request. All information received in response to this RFI or any follow-up information request that is marked Proprietary will be handled accordingly. Responses to the RFI will not be returned. At this time, questions concerning the composition and requirements for a future request for proposal will not be entertained.

7.0 DESCRIPTION: USAMRICD has a requirement for the following:

 In Vitro ADME Profiling on Five compounds provided by USAMRICD

Assays to Be Performed:

1) Aqueous Solubility at pH 7.4 (2% DMSO/PBS)

2) Stability in Physiological Fluids: Plasma, Simulated Gastric Fluid, Simulated Intestinal Fluid (One Concentration)

3) Stability in Liver Microsomes: 2 species, one concentration

4) Stability in Primary Human Hepatocytes, One Concentration

5) Inhibition of hERG in HEK293 Cells (Thalos® Ion Flux)

6) Plasma Protein Binding (Equilibrium Dialysis)

7) Parallel Artificial Membrane Permeation Assay

8) Blood Brain Barrier Parallel Artificial Membrane Permeation Assay

Results and Final Report:

Results of Assays 2, 3, and 4 will be reported as T1/2.

Results of 5 will be reported as IC50 or %inhibition at 10 uM. Results of 6 will be reported as % free, unbound.

Results of 7 and 8 will be reported as Papp (10-6 cm/sec) Results will be summarized in a report at the end of the study.

iv.            Rough order of magnitude (ROM) estimate of the cost to perform the scope of work

c. Brief summary of relevant experience pertaining to the requirement (for example government, academia or industry experience).
