The Centers for Disease Control and Prevention intends to issue a sole source purchase order to Roche Diagnostics, 9115 Hague Road, Indianapolis, IN 46250, for purchase of a MagnaPure LC 2.0 instrument, Automated Nucleic Acid Isolation System. This instrument will be used in the influenza branch for the following purpose: The wider international use of life-attenuated influenza vaccine based on donors of attenuation developed in Russia prompted WHO to request that influenza division of CDC prepare and quality control test LAIV vaccine reassortants for international use. Classical reassortment in hen’s eggs between WHO-recommended vaccine viruses and the viruses-donors of attenuation will be undertaken. This approach will result in large amount of clones, which would be screened for correct genetic formulation. To analyze genetic formulation of each clone, viral RNA should be isolated from each clone. In the process of the preparing of viral RNA from the virus clones there is a need of fast and automated nucleic acid isolation system with the reduction risk of contamination. Requirements are: fully automated, high-quality nucleic acid isolation, user-friendly software which controls all instrument functions as well as all steps of the purification protocols; the samples protected with a drop catcher, HEPA filter system, and UV decontamination; an automated nuclear acid isolation system which can process large quantities of samples in a relatively short time period (32 samples in 180 minutes or less); with the results being high-quality purified nucleic acids suitable for direct use in molecular biology applications such as PRC, RT-PCR, sequencing, and other array analysis to increase efficiency of extracted nucleic acid from virus clones which can be used in direct analysis for genetic composition. In addition, laboratory space is limited and the MagnaPure LC 2.0’s dimensions are ideal for this small space. CDC believes that this requirement is met by only one provider. This procurement will be processed under the authority of FAR 13.106-1(b)(1). Only one responsible source and no other sources will satisfy agency requirements. No solicitation is being issued.
