The National Institutes of Health (NIH) National Institute Diabetes and Digestive and Kidney Diseases (NIDDK) has a requirement to procure targated knock-in of a gene in the mouse genome. The goal of the work is to generate a mouse strain that carries a specific, heritable alteration in one of its genes. The task involves the application of gene targeting technology which is based on the process of homologous recombination to introduce the mutation into the desired gene. This is performed in mouse embryonic stem cells which are used to create founder mice carrying the targeted mutation. The mutation should be transmitted through the germline of the founder mice to allow generation of a colony of mice carrying the mutation for analysis. Specific requirements are: 1) Expertise in the field of gene targeting technology and a proven record of accomplishment. The provider should have been successful for a significant number of years in this field. Evidence for proven success includes the publication of a significant number of successful projects in reputable, peer-reviewed scientific journals. 2) Proven experience in performing similar projects in embryonic stem cell lines of different ,mouse strain origin. This includes the option of using C57BL/6- or 129 substrain-derived ES cells, each with high probabilities of success, supported by evidence from publications in peerìreviewed scientific journals. 3) Ability to perform the tasks within a realistic, competitive time frame for an important scientific project.
