Adding this cell sorter to the lab will greatly increase the lab's productivity and efficiency by not having to rely on reserving time at NIH flow cytometry cores. Currently limitation on sorting time does hold up some experiments and/or limit the timing when we can perform some experiments, which is critical since many of our experiments need to be carried out at specific embryonic ages and we do not have a whole lot of flexibility.
The benchtop SH800 cell sorter permits sorting of a wide range of cell sizes and applications using the 70um, 100um and 130um microfluidics sorting chips. Up to 4 lasers are available providing excitation lines of 405 nm, 488 nm, 561 nm and 638nm. Six freeform PMTs allow for the detection of fluorescence signals from any laser based on filter selection. This novel, chip-based design is fully integrated with comprehensive fluidics controls and advanced automation for set-up, acquisition, sort and analysis to make sorting less subjective, more precise and easier to use. System software is easy to learn and use. It supports sorting into tubes and 96- and 384-well plates via an optional Sort Deposition System. The software generates FCS 3.0 and FCS 3.1 files that also can be exported to third party analysis tools. Supports a variety of applications for individual and core labs. For applications where cross contamination is a concern, optional Electron beam irradiated consumables can be used to replace sample line and sorting chip.
Sony SH800 offers the preferred laser configuration (405/488/561nm). Similar machines sorters researched only offer the following 3-laser configuration setup (405/488/640nm) that is suboptimal for my needs. Sony has a small footprint which is important due to space limitations within the lab that has been allotted for this sorter. Sony sorter also uses a sorting chip, which will minimize running problems and required maintenance for the sorter.

The proposed contract action is for supplies or services for which the Government intends to solicit and negotiate with only one source under authority of the 41 U.S.C. 253(c) (1), FAR 6.302.
